AOD-9604 is a 16 amino acid piece of the tail end of human growth hormone, formally Tyr-hGH177-191. That carboxyl terminal region carries the fat mobilizing activity of the full hormone, and it leaves behind the growth signaling (Moré and Kenley, Journal of Endocrinology and Metabolism, 2014).

The animal work is the strongest part of the file. Heffernan and colleagues dosed obese mice chronically with AOD9604 and saw weight loss plus increased lipolytic sensitivity, along with a rise in beta-3 adrenoceptor expression that appears to amplify the effect (Endocrinology, 2001). Human results are thinner. Two oral phase IIb studies ran with 300 and 500 obese adults. Weight loss showed up in the earlier work and then went missing in the final study, which layered an intensive diet and exercise program on top of the drug. Across every human study reported, AOD9604 was well tolerated, and it left IGF-1 alone, which is the main way it separates from full growth hormone.

The FDA added AOD-9604 to Category 2 of its interim 503A bulks list on September 29, 2023, in the same batch as 18 other peptides including BPC-157 and Ipamorelin. That nomination was later withdrawn, and today AOD-9604 appears in the 'nominated but withdrawn' table on the FDA's bulk substances page while the agency continues evaluating it on its own initiative.
